How to Acclimate Tissue Culture Plants

A plant growing in tissue culture has spent its entire life in a carefully controlled environment.
Inside a vessel, humidity is nearly 100%, nutrients are supplied through agar, airflow is minimal, and environmental conditions remain stable. When that plant leaves the vessel, it suddenly faces a completely different world.
This transition is called acclimation.
Acclimating Tissue Culture Plants
For many tissue culture plants, acclimation is the most critical stage of cultivation. Done correctly, the plant adapts to life outside the flask and begins growing normally. Done too quickly, stress and dehydration can cause significant setbacks.
The goal is not speed.
The goal is helping the plant gradually learn to function in a natural environment.
Why Tissue Culture Plants Need Acclimation
Plants grown in culture develop differently than plants grown in open air.
Inside the vessel:
Humidity remains extremely high
Roots develop in agar rather than substrate
Leaves experience little airflow
Water loss is minimal
As a result, young plantlets often have thin leaves and underdeveloped protective structures that would normally help prevent dehydration.
Moving directly into typical household conditions can cause moisture loss faster than the plant can replace it.
Acclimation allows the plant to strengthen these systems gradually.
Step 1: Remove the Agar
The first step is carefully removing the plant from its culture vessel.
Any remaining agar should be gently rinsed away using clean water.
Why?
Residual agar can:
Encourage mold growth
Attract bacteria
Interfere with root establishment
Take care not to damage delicate roots during cleaning.
The goal is to leave the root system intact while removing as much media as possible.
Step 2: Choose an Appropriate Substrate
Different plants require different growing media.
Examples include:
Long-fiber sphagnum moss
Sphagnum/perlite mixes
Fine bark blends
Specialized tropical substrates
The best substrate is one that matches the species' natural preferences while providing adequate moisture and airflow.
Avoid heavily fertilized potting mixes unless the species specifically requires them.
Step 3: Start With High Humidity
Newly deflasked plants benefit from elevated humidity during the early stages of acclimation.
Common approaches include:
Humidity domes
Clear propagation containers
Covered trays
Controlled-environment cabinets
The purpose is not to keep humidity high forever.
Instead, it provides a temporary bridge between tissue culture conditions and the outside world.
Step 4: Reduce Humidity Gradually
This is where acclimation truly happens.
Over days or weeks, humidity should be reduced gradually by increasing airflow and ventilation.
For many species, this process may involve:
Opening vents
Cracking lids
Increasing air exchange
Removing covers incrementally
The exact timeline varies by species, plant size, and environmental conditions.
Patience often produces better results than rushing.
Step 5: Introduce Stronger Light Slowly
Freshly acclimating plants are often more sensitive to intense light than mature specimens.
Begin with:
Bright indirect light
Filtered greenhouse light
Moderate grow light intensity
As the plant establishes roots and begins producing new growth, lighting can gradually be increased.
New leaves produced during acclimation are often stronger and better adapted to normal growing conditions.
Signs of Successful Acclimation
Healthy acclimation is usually indicated by:
New root growth
New leaves
Improved coloration
Increased vigor
Stronger leaf structure
The most encouraging sign is often the appearance of growth that formed entirely outside the tissue culture vessel.
Once new growth begins, the plant has started adapting successfully.

Acclimation is where laboratory propagation becomes horticulture.
Every species responds differently, but the principles remain the same: remove agar carefully, provide appropriate substrate, maintain humidity initially, and reduce environmental support gradually.
